KIT DESCRIPTION:
Klebsiella pneumoniae is a Gram-negative, rod-shaped bacterium that is commonly found in the human gut but can cause severe infections when it spreads to other parts of the body. It is one of the most common pathogens associated with hospital-acquired infections, particularly in immunocompromised patients. K. pneumoniae is notorious for causing pneumonia, urinary tract infections, bloodstream infections, and wound infections. Over the years, this bacterium has developed significant antibiotic resistance, particularly through the production of extended-spectrum beta-lactamases (ESBLs) and carbapenemase enzymes, making it a major concern in healthcare settings. Its ability to acquire and transfer resistance genes highlights the need for ongoing research and effective infection control measures.
Positive Control: The positive control (PC) is a synthetic control in the kit containing 1x103 copies/µL of every target in the panel. Using a positive control from the same lot as the reagent kit is recommended.
Exogenous Control (EC): The Exogenous Control (EC) is a whole organism control containing 0.2x105 copies/µL of a non-pathogenic gram-positive bacterium. This control should be spiked into a sample volume ranging from 200 μL to 1 mL before the nucleic acid extraction, or extraction free processing. Detection of EC verifies the successful DNA extraction reaction and confirms the bacterial DNA extraction and amplification reaction.
Endogenous Control (RNaseP): Primers for RNAseP consistent expresses across different sample types, including human tissues and clinical specimens. Detection of RNAseP is indication of adequate sample collection quality, and PCR amplification. This internal control ensures the accuracy and reliability of diagnostic results by confirming the presence of human genetic material in the sample and validating the assay performance.
